Endoglucanases are key enzymes applied to the conversion of biomass aiming for second generation biofuel production. In the present study we obtained the small angle X-ray scattering (SAXS) structure of the G. trabeum endo-1,4-β-glucanase Cel12A and investigated the influence of an important parameter, temperature, on both secondary and tertiary structure of the enzyme and its activity. The CD analysis for GtCel12A revealed that changes in the CD spectra starts at 55 oC and the Tm calculated from the experimental CD sigmoid curve using the Boltzmann function was 60.

The purified domains were analyzed by circular dichroism spectroscopy, fluorescence spectroscopy, dynamic light scattering, and small-angle X-ray scattering, as well as with bioinformatics tools. Of the three major domains that comprise SEPT4, the N-terminal domain contains little regular secondary structure and may be intrinsically unstructured. The central GTPase domain is a mixed alpha/beta structure, probably based on an open beta sheet. As defined here, it is catalytically active and forms stable homodimers in vitro. The C-terminal domain also forms homodimers and can daniel dantas facebook be divided into two regions, the second of which is alpha-helical and consistent with a coiled-coil structure. These studies should provide a useful basis for future biophysical studies of SEPT4, including the structural basis for their involvement in diseases such as cancer and neurodegenerative disorders.

This paper presents a novel damage detection approach for Structural Health Monitoring (SHM) systems, which is based on Savitzky-Golay filter and time-domain analysis. In this technique, a specific PZT (actuator) excites the structure and three other ones are used as sensors to receive the structural responses. This method is quite similar to Lamb wave based methods, but the excitation procedure and the analysis of the response signals are different. For the proposed method, a wide band chirp signal, with low amplitude and variable frequency, is used to excite the structure.
